Alpha®: Amplified Luminescent Proximity Homogeneous Assay Bead-based - Donor and Acceptor Beads Homogeneous - no wash steps Versatility - Detects virtually any molecule from large endogenous protein complexes to very small peptides Works in a variety of sample types including serum, plasma, cell lysates and cell supernatant High sensitivity (fmol) - Amplified signal Low background - Emission wavelength is lower than Excitation Based on AS. Bead based luminscent assay. Donor beads and Acceptor beads. When binding partners bring these 2 beads into close proximity there can be transfer of E between the beads. Upon laser excitation the donor bead produces a oxygen singlet that can excite the Acceptor bead which then emit light at 520-620nm Advantage of AS: large distance between beads (up to 400nm) allows measurement of intereactions between large molecules.

Beads conjugated with protein A, G and various anti-species antibodies allows the capture of primary antibodies through their heavy chains whereas, Protein L coated beads allows the capture antibody light chains.

Cellular assay for measuring p65-IkBa interaction Protein-protein interactions could be also studied in a cellular context. The example presented on this slide was developped by PerkinElmer R&D scientists using AlphaLISA reagents. The assay aimed at measuring the interaction prevailing between IkBa and p65 in Hela cells. Using antibodies specific to IkBa and p65 epitopes non-overlapping with their binding interface, AlphaLISA assays were developed to measure the IkBa/p65 complex and IkBa or p65 alone in cellular extracts. After treating Hela cells with increasing dose of TNFa, one can measure the concomitant disappearance of the IkBa/p65 complex and IkBa due to the degradation of the latter protein. Levels of p65 remained unchanged. The assay also allowed to measure the kinetics of IkBa/p65 disappearance: as reported by other groups using traditional Western Blotting technique, the phenomenon is transitory and IkBa/p65 is reformed after one hour following the accumulation of newly available IkBa.
