Mais ce n’est pas la séquence primaire qui est cause de cette asymétrie ce serait un encombrement stérique due à la liaison de la partie N terminale. Lambda se fixe sur du straight DNA ce n’est pas la conformation qui change la spécificité. Lambda et Cro ont une reconnaissance par lecture de la séquence (readout reconnaissance des bases) Indirect readout reconnaissance du backbone phosphate 3 points important pour La reconnaissance 1-Déformation de l’ADN (important pour EcoRV et BamHI 2-Coopérativité 3-Effet quantitatif d’une séquence spécifique ou non Les différences ne sont pas dues à la géométrie de la cassure mais plutôt à l’énergie mise en cause pour obtenir cette cassure (CAP Lawson 2004). Le point déterminant dans la géométrie locale de l’ADN serait les interaction Prot/ADN plutôt que la séquence seule. Augé gouillou 2005 MCB les deux demisites garde une activité de fixation de la tnp de 3% en EMSA Bigot 2005 sur l’ADN nu du 3’ ona un angle de 20° Fixation asymétrique du dimère de transposase 1/2 site différent ?
